A breakthrough in the usage of Resveratrol
« on: May 08, 2009, 12:10:46 AM »
Hello everyone

Today i attended Professor R. Gambari lecture about induction of fetal HB. It was so interesting and very promising.

He talked about different points that i will discuss later but what i consider a very important outcome is his study of resveratrol offered by different brand names that are already found in the market.

Professor Gambari had discovered that there are certain additives that are added by drug companies that are found to inhibit the effect of resveratrol in the induction of fetal Hb

Quote
Since resveratrol is already present in the pharmaceutical market as antioxidant, we have undertaken a study to verify erythroid induction using resveratrol formulations obtained from different companies. This is a crucial investigation, since resveratrol is usually employed together with other additives, such as curcumin, extracts from grape seeds, vitamins. We have demonstrated that several commercially-available resveratrol-based formulation contain additives inhibiting erythroid differentiation; the best formulations for HbF production by beta-thal cells are those containing the highest proportion of resveratrol.

Actually i don't have the lecture copied, but he mentioned what are the components that increase and decrease the resveratrol efficency. The best Brand name that was proved to have 100% effiecency is resveratrol from Transmax Biotivia and also the brand Longenviex

To my surprise (because most of us use it) he found that resveratrol from the brand Puritan Pride was one of the worest resveratrols in terms of effiency. It is efficent by only 3% in the induction of fetal HB. He insisted so much that resveratrol should be taken from a brand that does not add additives to the resveratrol

I guess that this finding is so important cause we can be using an inefficent brand and therefore can not measure or achieve any benefits from these natural inducers

To my surprise too, in his lecture graphs showed that the usage of these natural inducers were able to increase the fetal Hb more than the hydroxyurea

I asked him about the Nature's Way brand that i use, he said that he did not test it but i am happy to say that he took some capsules from me to test the efficency of resveratrol in this particular brand and will have an answer within 20 days

Professor Gambari is so much interested in the natural HB inducers and i told him about the Aloe vera that i am giving to my son and also about the informal study of Carao that we are doing here through our group, he took the names and told me that he will test them on different kind of  mutation and their ability to induce fetal HB.

He was so interested in knowing our observation after using carao and is ready to investigate in it more through his lab so may be it will be a good idea if Lloyd Standish and Professor Gambari would get into contact to conduct a more accurate and specialised study in evaluating the efficency of Carao too.

I hope that this helps and that this important note would e taken in consideration to anyone using resveratrol

Manal,

Do you have a list of these additives?

There are very few additives in most brands of Resveratrol, including Puritan, and what is there are the same additives that are present in many supplement products. What should we be looking for that can alter the effect of the supplements? Also, Puritan does have more than one source of resveratrol for their products. I would like to know if there is any difference between resveratrol made from grapes and resveratrol derived from Japanese knotweed (Polygonum cuspidatum).

All we are saying is give thals a chance.

Andy,

Actually there was a list of the componenets that increase and decrease the efficency of resveratrol in inducng the fetal Hb, but i wasn't able to copy them cause they slides were moving quickly. I will try my best to get the lecture on a CD if this is allowed.

As for your other questions, i really don't have an answer for them but i will forward them to Professor Gambari and let you know his answer. If you would like me to send you his email too let me know

Manal

PS: one of the things that caught my atttention is the huge difference in price between resveratrol from the Transmax Biotivia (the brand with the most efficent resveratrol. costs $74 ) and the other resveratrols from different brands that have the average of $7 t0 $10

I am glad that you guys has some things left  that can help too (a bit ( blood transfusion)
I am looking forward to hear his results!!

Best wishes, Dore

Hello Andy

Professor Gambari has answered your question and the answer was as follows

Quote
1- Is there a list of additives that we should avoid when using resveratrol or other supplements?? There are very few additives in most brands of Resveratrol, and what is there are the same additives that are present in many supplement products. What should we be looking for that can alter the effect of the supplements or resveratrol?
We noted that some additives inhibit erythroid differentiation and HbF production. One example is quercetin, another example is skin grape extracts. In addition, we noticed that the best products are those that indicate resveratrol at 99% concentration. As soon as we complete the experiment, you will be informed about the full list of compounds possibly interfering with resveratrol activity. Please note that all the information we have are not IN VIVO, but IN VITRO on cells from beta-thalassemia patients.

2- Also, some resveratrol brands have more than one source of resveratrol for their products. I would like to know if there is any difference between resveratrol made from grapes and resveratrol derived from Japanese knotweed (Polygonum cuspidatum).??
In our hands resveratrol from grapes and from P. cuspidatum are both active.

Regarding Professor Gambari answer vs. David Sinclair for Longevinex saying that Quercetin is essential to the effectiveness of the Longevinex in that it "...quercetin also helps to activate the same gene that resveratrol does, albeit less potently".  I thought Pro. Gambari said that quercetin inhibit the growth of HbF.  So if there are no HbF production doesn't that mean the present of resverotrol are minuscule to non?  See a thread I found below.  Maybe you could make sense of what they are trying to say.

QUOTE
"....Sinclair found that only research-grade resveratrol produced under hypoxic conditions and sealed under nitrogen during storage were found to have significant biological activity. The compound simply oxidizes rapidly... think of a sliced apple which turns brown after a short time. Longevinex is the first supplement to be produced under a nitrogen environment, and sealed in an air-tight capsule with a nitrogen bubble inside to protect from oxidation. For the same reason, the resveratrol in red wine is quickly oxidized in less than a day after the bottle is opened. Since wine can be purchased in the plastic collapsable bags, some have chosen to supplement with resveratrol in this fashion. However, one would have to drink over 10 glasses of typical red wine per day to get the same amount of resveratrol, if my memory serves; although some red wines contain more resveratrol than others.

Another problem with resveratrol is that it rapidly forms conjugates both in the digestive tract and during the initial pass through the liver. Quercetin can saturate and bind to the same compounds which conjugate with resveratrol, so Longevinex also includes quercetin in the pill. However, it is a small amount, and my guess is that all of the quercetin, plus all of the resveratrol in the pill will become conjugated. A possible solution to this is to take extra quercetin (say 500mg) shortly before taking Longevinex. Two published studies now have found that resveratrol rapidly forms conjugates and is likely not bioavailable. Quercetin is believed to inhibit this conjugation, and quercetin also helps to activate the same gene that resveratrol does, albeit less potently."

But does this oxidized or conjugated resveratrol work as an antioxidant, or has it any effect on SIRT gene that is supposed to work like a low-calorie diet when activated by resveratrol? Maybe there is at least antioxidant effect when there is no fetal hb inducing effect (although I doubt it)?

The most efficient way of administering resveratrol in humans appears to be buccal delivery, that is without swallowing, by direct absorption through the inside of the mouth. When one mg of resveratrol in 50 mL solution was retained in the mouth for one min before swallowing, 37 ng/ml of free resveratrol were measured in plasma two minutes later. This level of unchanged resveratrol in blood can only be achieved with 250 mg of resveratrol taken in a pill form.
